In the 72-week, phase 3 ATTAIN-2 trial, researchers evaluated once-daily orforglipron—a novel oral, small-molecule GLP-1 receptor agonist—in 1,613 adults with type 2 diabetes and obesity or overweight. Participants were randomized to receive 6 mg, 12 mg, or 36 mg of orforglipron, or a placebo, alongside lifestyle modifications. By week 72, those taking the highest dose (36 mg) achieved a significant mean weight reduction of 9.6% compared to just 2.5% in the placebo group. The oral medication also dramatically improved blood sugar levels (HbA1c) with manageable, primarily gastrointestinal side effects.
Why this is interesting
Currently, most highly effective GLP-1 therapies require weekly injections, which can be a barrier for many patients. Orforglipron is a daily pill, not an injection, representing a major leap forward in convenience and accessibility. Because it is a "small-molecule" drug rather than a peptide, it is easier to manufacture and take orally. For patients living with both type 2 diabetes and obesity, this study proves that a simple daily pill can deliver double-digit weight loss and robust blood sugar control without the need for needles.
