The LACI-2 clinical trial evaluated 363 adults who had experienced a lacunar stroke, a type of stroke caused by cerebral small vessel disease. Researchers tested two established medications, isosorbide mononitrate and cilostazol, individually and in combination. By six months, patients taking isosorbide mononitrate had fewer cardiovascular and dependency events, while those on cilostazol showed better cognitive outcomes. Most notably, combining both drugs yielded rapid, significant benefits in patients' cognition, mood, and overall quality of life, proving that positive effects begin much earlier than previously confirmed.
Why this is interesting
Lacunar strokes cause small but cumulative damage, often leading to vascular dementia, and there are currently few targeted treatments to halt this progression. This study demonstrates that repurposing two inexpensive, widely available cardiovascular drugs can actively protect brain function and mood within just six months.
